if it had to be one place in the us, it would have to be the hometown dc or the dmv would be epic, with such a vastly diffferent amount of terrian and the beltway my god that would be epic

Vegas, Seattle and Chicago are all good choices.  I was always disappointed by the recreation of Vegas in SA and think they need to 're-do' it like they did with LC in GTA IV, I think it would offer some interesting dynamics with the casino, tourist and mob culture, and the desert area around it would of course come into play.  I can imagine them spending almost all of their time on recreating the strip, which would probably make for some really amazing visuals and immersion.

They should make a GTA in the Netherlands. Most storylines have drugs involved or are the main cause of all problems. So, that would define to the drug wars/hunting/enforcing.

hmm i think somewhere in Canada would be good, British Columbia is quite a popular place, has a bit of everything, gangs, drugs, the coast lol since thats how drugs are imported, or even Nova Scotia, Al Capone used Halifax's Harbours for importing illegal goods, why couldn't R* use it?

Remember: They need a fake name for every location. That'd rule out "the whole US".
